Who they are
Titania's grievance with Jennifer Walters is trademark law: she filed "She-Hulk" first and intends to keep it. The series uses her to make its actual point — that being a woman with powers in public is mostly a branding problem — and gives her the single best pratfall in the MCU.

Made on Battleworld
Mary MacPherran was a bullied woman from a Denver trailer park who Doctor Doom turned into a powerhouse during the first Secret Wars, and she has spent forty years being the Hulk family's most persistent grudge. Her rivalry with She-Hulk in print is genuinely violent rather than litigious.
